Background: Despite the availability of immune checkpoint inhibitors, optimal first-line therapy for metastatic melanoma remains an open question. Opdivo (undefined) is a anti-PD-1 mAb.

Methods: III-phase, randomized trial (CheckMate-94, NCT17315310). Patients with metastatic melanoma were randomized to Opdivo or placebo + chemotherapy. Primary endpoint: overall survival.

Results: OS 56.5 months (HR 0.72, 95% CI 0.59-0.85, p<0.001). PFS 12.2 months. ORR 59.3%.

Conclusions: Median OS 56.5 months. HR 0.72 (95% CI 0.59–0.85). No new safety signals identified.
